Biological Background: MEPCE Function and Localization
- MEPCE (7SK snRNA methylphosphate capping enzyme), also known as Bicoid-interacting protein 3 homolog (Bin3 homolog), is a S-adenosyl-L-methionine-dependent methyltransferase.
- It catalyzes the addition of a methylphosphate cap to the 5'-end of 7SK snRNA, stabilizing this non-coding RNA. Related references: PMID:17643375, PMID:19906723, PMID:30559425
- MEPCE has a non-enzymatic function as a core component of the 7SK ribonucleoprotein (RNP) complex, which sequesters P-TEFb to inhibit RNA polymerase II phosphorylation and transcriptional elongation. Related references: PMID:17643375
- The 7SK RNP complex also promotes snRNA gene transcription by interacting with the little elongation complex (LEC). Related references: PMID:28254838
- MEPCE is essential for 7SK RNA stability and 7SK RNP assembly; interaction with LARP7 occludes its catalytic center. Related references: PMID:19906723, PMID:38100593
- Additionally, MEPCE is required for the stability of U6 small nuclear RNAs. Related references: PMID:38100593
- The protein localizes to the nucleus and is expressed tissue-specifically in chronic myeloid leukemia cells, adrenal gland, brain, cerebellum, kidney, lung, mammary gland, and testis. Related references: PMID:12358911
- MEPCE undergoes post-translational modifications including acetylation, methylation, phosphorylation, and ubiquitin-like conjugation, and is subject to alternative splicing.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 470-689 at the C-terminus of human MEPCE, a region that may be present in full-length and certain alternatively spliced isoforms.
- For immunofluorescence/immunocytochemistry (IF/ICC), optimization of fixation and permeabilization protocols is recommended due to nuclear localization of the target.
- ELISA assays may require appropriate controls and dilution optimization for specific sample types.
- Consider validating the antibody performance in your relevant cell lines or tissues, particularly those with known MEPCE expression.
