Biological Background: MEP1A (Meprin A Subunit Alpha) Function and Localization
- MEP1A encodes the alpha subunit of meprin A, a zinc-dependent metalloprotease belonging to the astacin family of proteases.
- The protein is also known as endopeptidase-2, PABA peptide hydrolase (PPH alpha), and N-benzoyl-L-tyrosyl-P-amino-benzoic acid hydrolase subunit alpha.
- Meprin A is a membrane-bound enzyme, anchored to the cell surface via a single-pass transmembrane domain.
- The precursor form is a zymogen that requires proteolytic removal of the pro-peptide for catalytic activation.
- Structurally, MEP1A contains an EGF-like domain, multiple disulfide bonds, and is heavily glycosylated, existing as a proteoglycan.
- Based on UniProt annotation, key features include hydrolase activity, metal-binding, and zinc coordination, with a molecular weight of approximately 84.4 kDa.
Experimental Guidance and Technical Tips
- For Western blotting, consider using 8–10% SDS-PAGE to optimize separation of the ~84 kDa MEP1A protein.
- As a membrane protein, efficient extraction may require detergents such as Triton X-100 or NP-40 in the lysis buffer; validate conditions in your sample system.
- When performing ELISA, titrate both coating antigen and primary antibody to establish optimal working concentrations; antigen may be recombinant MEP1A or cell/tissue lysates.
- The antibody is reported to cross-react with mouse and rat MEP1A; however, always confirm cross-reactivity in your specific tissue or cell types before routine use.
- Because this is a polyclonal antibody, lot-to-lot variability should be assessed and controlled in critical quantitative assays.
