- MEK7 (also known as MKK7, JNKK2, SAPKK4) is a dual-specificity mitogen-activated protein kinase kinase essential for the stress-activated protein kinase/c-Jun N-terminal kinase (SAP/JNK) signaling pathway.
- Together with MKK4, MEK7 directly activates JNK1, JNK2, and JNK3 by phosphorylation, with a preference for the Thr residue in the Thr-Pro-Tyr activation motif, triggering JNK activity.
- The MKK7/JNK pathway is activated by pro-inflammatory cytokines and plays a critical role in cellular stress responses, inflammation, and apoptosis.
- MEK7 participates in mitochondrial death signaling, promoting cytochrome c release and leading to programmed cell death.
- It functions in a non-canonical MAPK pathway with MAP3K12 and ERK1/2 to enhance AP-1-mediated transcription of APP in response to APOE. Related references: PMID:28111074
- Subcellular localization: found in both the nucleus and cytoplasm.
- Widely expressed in human tissues, with highest levels in skeletal muscle; isoform 3 is present at low levels in placenta, fetal liver, and skeletal muscle.
