Biological Background: MEK7 Function and Localization

  • MEK7 (also known as MKK7, JNKK2, SAPKK4) is a dual-specificity mitogen-activated protein kinase kinase essential for the stress-activated protein kinase/c-Jun N-terminal kinase (SAP/JNK) signaling pathway.
  • Together with MKK4, MEK7 directly activates JNK1, JNK2, and JNK3 by phosphorylation, with a preference for the Thr residue in the Thr-Pro-Tyr activation motif, triggering JNK activity.
  • The MKK7/JNK pathway is activated by pro-inflammatory cytokines and plays a critical role in cellular stress responses, inflammation, and apoptosis.
  • MEK7 participates in mitochondrial death signaling, promoting cytochrome c release and leading to programmed cell death.
  • It functions in a non-canonical MAPK pathway with MAP3K12 and ERK1/2 to enhance AP-1-mediated transcription of APP in response to APOE. Related references: PMID:28111074
  • Subcellular localization: found in both the nucleus and cytoplasm.
  • Widely expressed in human tissues, with highest levels in skeletal muscle; isoform 3 is present at low levels in placenta, fetal liver, and skeletal muscle.
  • MEK7 is regulated by phosphorylation and acetylation, and belongs to the protein kinase superfamily with an ATP-binding site and serine/threonine/tyrosine kinase activity.

Experimental Guidance and Technical Tips

  • This antibody is reported as knockout (KO) validated, confirming its specificity for MEK7; use appropriate KO cell or tissue lysates as negative controls in Western blotting.
  • The immunogen (aa 1–250) targets the N-terminal region of human MEK7, which is conserved across isoforms; expect detection of the full-length protein (~47 kDa) and potentially splice variants in WB.
  • For Western blot, consider a dilution range of 1:500–1:2000, using standard blocking and detection protocols, and validate in human, mouse, or rat cell lines expressing MEK7.
  • In IF/ICC, permeabilization and fixation conditions should be optimized; include a no-primary control to assess non-specific binding.
  • ELISA performance relies on coating antigen purity; pair with a validated capture antibody if developing a sandwich assay.
