Biological Background: MEK7 Function and Localization

  • Dual specificity mitogen-activated protein kinase kinase 7 (MAP2K7), also known as MEK7, MKK7, JNKK2, SAPKK4, PRKMK7, is an essential component of the stress-activated protein kinase/c-Jun N-terminal kinase (SAP/JNK) signaling pathway. It belongs to the MAP kinase kinase family and acts as a critical upstream activator of JNKs (MAPK8, MAPK9, MAPK10).
  • Together with MAP2K4/MKK4, MAP2K7 is one of the only known kinases that directly phosphorylate and activate JNKs. While MKK4 preferentially phosphorylates the Tyr residue in the Thr-Pro-Tyr activation motif of JNKs, MKK7 shows a strong preference for the Thr residue. Monophosphorylation of JNKs on Thr by MKK7 is sufficient to increase JNK activity, with Tyr phosphorylation by MKK4 ensuring optimal activation.
  • MAP2K7 plays a specific role in mediating JNK signal transduction triggered by pro-inflammatory cytokines. The MKK/JNK signaling pathway also participates in mitochondrial death signaling, promoting cytochrome c release and subsequent apoptosis.
  • Part of a non-canonical MAPK signaling pathway, involving upstream MAP3K12 kinase and downstream MAPK1/ERK2 and MAPK3/ERK1, that enhances AP-1-mediated transcription of the amyloid precursor protein (APP) in response to APOE. Related references: PMID:28111074
  • Predominantly localizes in the nucleus and cytoplasm, consistent with its role in transmitting extracellular stress signals to nuclear transcriptional responses.
  • Ubiquitously expressed, with highest levels in skeletal muscle. Isoform 3 is found at low levels in placenta, fetal liver, and skeletal muscle.
  • Post-translationally regulated through acetylation and phosphorylation; classified as a serine/threonine-protein kinase and a tyrosine-protein kinase, requiring magnesium ions as cofactors for ATP binding and kinase activity.

Experimental Guidance and Technical Tips

  • The immunogen used is a synthetic peptide spanning amino acids 320–419 of human MEK7 (UniProt O14733). For Western blot, expect a band at approximately 47 kDa; consider including appropriate positive controls such as HeLa or HEK293 cell lysates to confirm reactivity.
  • This antibody is validated for WB and ELISA. For ELISA, optimal coating concentration and detection conditions should be empirically determined; consider testing 1–10 µg/mL as a starting range for capture antibody.
  • Species cross-reactivity is reported for human, mouse, and rat. When using with samples from these species, verify performance in your experimental system, as sequence homology may affect sensitivity.
