  • MEK5, also known as MAP2K5, is a dual specificity mitogen-activated protein kinase kinase that can phosphorylate both serine/threonine and tyrosine residues.
  • It acts as a scaffold for the formation of a ternary signaling complex involving MAP3K2/MAP3K3, MAP3K5, and MAPK7 (ERK5), playing a key role in the MAPK/ERK signaling cascade.
  • Activation of this pathway is critical for protecting cells from stress-induced apoptosis, promoting neuronal survival, and supporting cardiac development and angiogenesis.
  • In cardiomyocytes, MEK5 functions as a negative regulator of apoptosis by facilitating STUB1/CHIP-mediated ubiquitination and degradation of ICER-type isoforms of CREM (By similarity).
  • The protein is broadly expressed in adult tissues, with particularly high abundance in heart and skeletal muscle.

  • For Western blotting, the expected band size is approximately 50 kDa; validate with appropriate positive and negative controls.
  • In immunohistochemistry and immunofluorescence, heart and skeletal muscle tissues are suitable positive controls given the high expression of MEK5.
