Biological Background: MEK1/MEK2 Function and Localization

  • MEK1 (MAP2K1) is a dual-specificity protein kinase that plays a central role in the mitogen-activated protein kinase (MAPK) signaling cascade, specifically in the ERK pathway. Upon activation by RAF1, it phosphorylates both threonine and tyrosine residues in MAPK3/ERK1 and MAPK1/ERK2.
  • MEK1 and its close homolog MEK2 (MAP2K2) are functionally overlapping and cooperate to transduce signals from cell surface receptors to nuclear and cytoplasmic effectors, regulating diverse processes including cell proliferation, differentiation, survival, and adhesion.

Experimental Guidance and Technical Tips

  • Since this polyclonal antibody is raised against an internal peptide of human MAP2K1, it may detect both MEK1 and MEK2 due to sequence similarity, as well as mouse and rat orthologs. Validate specificity in your model system.
  • For Western blotting, using reducing conditions and whole-cell lysates, expect bands around 43-44 kDa for MEK1 and 45 kDa for MEK2; additional bands may reflect post-translational modifications or degradation products.
  • For ELISA, the antibody can be used as capture or detection reagent; titrate and optimize for direct or indirect assay formats.
  • Include appropriate positive and negative controls (e.g., stimulated vs unstimulated cells) to confirm pathway-dependent detection.
