Biological Background: MEK1 Function and Localization
- MEK1 (MAP2K1) is a dual-specificity kinase that phosphorylates both threonine and tyrosine residues in the activation loop of ERK1/2 (MAPK3/1), a critical step in the MAPK/ERK signaling cascade.
- Acts downstream of RAS-RAF1 and is essential for transmitting signals from cell-surface receptors to intracellular effectors, thereby regulating processes such as cell proliferation, differentiation, and survival.
- MEK1 activates BRAF through a KSR1/KSR2-dependent mechanism, releasing BRAF autoinhibition and enhancing signal propagation. Related references: PMID:29433126
- Subcellular localization spans the cytoplasm, centrosome, spindle pole, nucleus, and membrane, reflecting its roles in mitosis (Golgi fragmentation), endosomal trafficking, and nuclear export (e.g., PPARG).
- Widely expressed in human tissues, with notably low levels in the brain. Disease-associated mutations are linked to cardiomyopathy, ectodermal dysplasia, and intellectual disability.

Experimental Guidance and Technical Tips
- The immunogen used was a synthetic peptide corresponding to amino acids 1–100 of human MEK1. Epitope mapping suggests recognition of the N-terminal region, which is well-conserved; however, cross-reactivity with MEK2 should be experimentally excluded if specificity is critical.
- For Western blotting, a band around 43 kDa is expected. Validate using a positive control lysate from human, mouse, or rat cell lines known to express MEK1 (e.g., HeLa, NIH/3T3, PC12).
- For immunofluorescence/ICC, optimize fixation/permeabilization conditions, as MEK1 can be cytosolic, membrane-associated, or nuclear depending on cell state.
- For immunoprecipitation, use a gentle lysis buffer to preserve protein interactions. Consider cross-linking if co-IP of MAPK pathway components is desired.
- ELISA applications may employ the antibody for detection of recombinant or endogenous MEK1; titrate and include appropriate controls.
