Biological Background: MEK1 Function and Localization
- MEK1 (dual specificity mitogen-activated protein kinase kinase 1) is an essential component of the MAPK/ERK signal transduction cascade, relaying signals from RAF1 to ERK1/2 upon extracellular ligand stimulation.
- Catalyzes the concomitant phosphorylation of threonine and tyrosine in the Thr-Glu-Tyr motif of ERK1/MAPK3 and ERK2/MAPK1, leading to their activation. This pathway regulates cell growth, adhesion, survival and differentiation.
- Activates BRAF in a KSR1/2-dependent manner by promoting KSR-BRAF dimerization and BRAF activation (PubMed:29433126). Related references: PMID:29433126
- Localizes to cytoplasm, centrosome, spindle pole body, membrane, and nucleus, highlighting a role in mitotic Golgi fragmentation and endosomal dynamics.
- Widely expressed across tissues with extremely low levels in the brain; implicated in cardiomyopathies, ectodermal dysplasia, and intellectual disability based on disease-associated variants.
- Post-translational modifications include acetylation and phosphorylation; classified as a serine/threonine and tyrosine dual-specificity kinase.
Experimental Guidance and Technical Tips
- For Western blotting, a band near 43 kDa may be observed; validate detection in your specific sample system.
- For immunohistochemistry/immunofluorescence, consider antigen retrieval and appropriate controls due to dual cytoplasmic and nuclear localization.
- Given cross-reactivity with mouse and rat, this antibody may be suitable for model organism studies; confirm specificity and optimize conditions.
- The immunogen targets the N-terminal region (aa 1–100); note that some splice variants or interacting partners might influence epitope accessibility.
