Biological Background: MEIS2 Function and Localization
- Homeobox protein Meis2 (also known as Meis1-related protein 1, MRG1) is a transcription factor encoded by the MEIS2 gene. Alternative splicing produces multiple isoforms with distinct functions.
- Meis2 binds to HOX or PBX proteins to form dimers, or to a DNA-bound dimer of PBX and HOX, stabilizing the homeoprotein-DNA complex. Isoform 3 is required for transcriptional activation of the ELA1 enhancer in pancreatic acinar cells in complex with PDX1:PBX1b.
- Isoforms 3 and 4 can bind to an EPHA8 promoter sequence containing the motif 5'-CGGTCA-3' and, in cooperation with PBX proteins, are proposed to activate EPHA8 transcription in the developing midbrain.
- Meis2 can bind the DNA sequence 5'-TGACAG-3' in the activator ACT sequence of the DRD1 promoter and activate transcription; however, isoform 5 cannot activate DRD1.
- Subcellular localization: primarily nuclear, but also detected in the cytoplasm and perinuclear region.
- It is widely expressed, with high levels in lymphoid organs of hematopoietic tissues and in specific brain regions such as the putamen.
- Meis2 is implicated in developmental processes and is associated with disease variants, including intellectual disability.
Experimental Guidance and Technical Tips
- For Western blotting, use appropriate positive and negative cell/tissue lysates. A band around 52kDa is expected, but alternative isoforms or modifications may produce additional bands.
- Since this is a polyclonal antibody raised against a recombinant fragment (aa 351-470), it may recognize multiple MEIS2 isoforms. Verify specificity using relevant controls.
- The antibody shows cross-reactivity with mouse and rat. Validate performance in these species with appropriate sample preparation.
- For ELISA, optimize coating antigen concentration and antibody dilution using the immunogen or full-length protein as standard.
- Consider including a nuclear extract as a positive control to enrich for MEIS2 detection.
