Biological Background: Homeobox protein Meis1 Function and Localization
- MEIS1 encodes homeobox protein Meis1, a transcription factor involved in development and hematopoiesis.
- It acts as a transcriptional regulator of PAX6 and as a cofactor for HOXA7 and HOXA9, contributing to myeloid leukemia induction.
- In complex with PBX1 or PBX2, MEIS1 activates PF4 transcription, and is required for megakaryocyte lineage development and vascular patterning.
- The protein localizes to the nucleus and contains a DNA-binding homeobox domain.
- MEIS1 is expressed at low levels in normal immunohepatopoietic tissues, including fetal liver, but shows elevated expression in certain myeloid leukemia cell lines, especially those with megakaryocytic-erythroid phenotype, and in the cerebellum.
- Alternative splicing produces multiple isoforms, and the protein is subject to post-translational modifications.
Experimental Guidance and Technical Tips
- This antibody was raised against a synthetic peptide corresponding to amino acids 300-390 of human MEIS1; consider that this region is predicted to be accessible in the native protein based on sequence analysis.
- For Western blotting, a band around 43 kDa is expected; optimize primary antibody dilution starting at 1:500-1:2000 and validate with positive controls such as leukemia cell lines or cerebellar tissue lysates.
- In immunofluorescence/immunocytochemistry, nuclear staining should be observed; include appropriate fixation and permeabilization steps.
- For ELISA, the peptide immunogen can serve as a positive control; assay conditions should be optimized individually.
- Cross-reactivity with mouse and rat MEIS1 is expected due to sequence homology; however, confirm specificity in your target species by sequence alignment.
