Biological Background: MEF2A Function and Localization

  • MEF2A is a member of the myocyte enhancer factor-2 (MEF2) family of transcription factors, characterized by a MADS-box and MEF2 domain.
  • It binds specifically to the MEF2 element (5'-YTAATTAR-3') in the regulatory regions of numerous muscle-specific, growth factor-induced, and stress-responsive genes.
  • MEF2A plays critical roles in skeletal and cardiac muscle development, as well as in neuronal differentiation and survival.
  • Through p38 MAPK signaling, MEF2A regulates cell growth, survival, and apoptosis in a tissue-specific manner.
  • Alternative splicing produces multiple isoforms; isoform MEF2 and MEFA are predominantly expressed in skeletal muscle, cardiac muscle, and brain, while RSRFC4 and RSRFC9 are ubiquitously expressed.
  • MEF2A localizes to the nucleus and undergoes various post-translational modifications, including phosphorylation, acetylation, sumoylation, and ubiquitin-like conjugation.
  • In cerebellar granule neurons, sumoylated and phosphorylated MEF2A represses NUR77 transcription, promoting synaptic differentiation.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the N-terminal region (aa 1-100) of human MEF2A, which is conserved across human, mouse, and rat, consistent with the reported cross-reactivity.
  • For Western blotting, the predicted molecular weight is approximately 55 kDa; however, post-translational modifications may result in observed bands at shifted positions. Consider using appropriate positive control lysates from skeletal muscle, heart, or brain tissues.
  • In immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), nuclear staining is expected. Antigen retrieval and permeabilization steps should be optimized for the specific tissue or cell type.
  • For ELISA, pairing with a matched detection antibody may be required; consider evaluating the antibody in a sandwich ELISA format if quantitative detection is desired.
  • Always validate the antibody in the relevant sample system and include appropriate controls, as performance may vary depending on the experimental conditions.
