- MED6 (Mediator of RNA polymerase II transcription subunit 6) is a component of the Mediator complex, a multiprotein coactivator required for regulated transcription of nearly all RNA polymerase II-dependent genes.
- It functions as a molecular bridge, conveying regulatory signals from gene-specific activators to the basal transcription machinery, facilitating the assembly of the preinitiation complex.
- The protein is predominantly localized to the nucleus, consistent with its role in transcription regulation.
- MED6 is also known under alternative names such as Activator-recruited cofactor 33 kDa component (ARC33), Mediator complex subunit 6 (hMed6), and Renal carcinoma antigen NY-REN-28.
- Alternative splicing of the MED6 gene produces multiple transcript variants, potentially contributing to functional diversity.
- Post-translational modifications include acetylation and isopeptide bond formation (ubiquitination), which may modulate its activity or interactions.
Experimental Guidance and Technical Tips
- The antibody is raised against residues 27–246 of human MED6, encompassing the core region. It is expected to detect both full-length MED6 and potential splice variants containing this domain, if expressed.
- For Western blot, a band corresponding to approximately 28 kDa (calculated molecular weight: 28.4 kDa) is expected. Post-translational modifications such as acetylation and ubiquitination may cause minor band shifts.
- This antibody is validated for ELISA and Western blot. It may also be suitable for other applications (e.g., immunoprecipitation, immunofluorescence); however, these should be optimized and verified experimentally.
- Cross-reactivity is reported for human. If studying other organisms, validation in the relevant sample system is advised.
- As a polyclonal antibody, it provides high sensitivity but may exhibit batch-to-batch variation. Including appropriate positive and negative controls is recommended, such as cell lines with known MED6 expression levels or siRNA-mediated knockdown.
