Biological Background: MECR Function and Localization
- MECR (NRBF1/ETR1) functions as a mitochondrial enoyl-[acyl-carrier-protein] reductase, catalyzing the NADPH-dependent reduction of trans-2-enoyl thioesters in fatty acid synthesis type II. It accepts both ACP and CoA thioester substrates in vitro, with a preference for medium-chain fatty acids. Related references: PMID:12654921 PMID:18479707 PMID:27817865
- The enzyme may supply the octanoyl chain for lipoic acid biosynthesis, thereby regulating protein lipoylation and mitochondrial respiratory activity, notably in Purkinje cells.
- MECR is involved in iron homeostasis; it affects Fe-S cluster assembly and ceramide metabolism, and is required for maintaining proper mitochondrial morphology and bioenergetic functions. Related references: PMID:37653044
- It is localized to mitochondria, cytoplasm, and nucleus.
- Highly expressed in skeletal and heart muscle; lower expression in placenta, liver, kidney, pancreas; weak or absent in lung.
- The protein undergoes acetylation and alternative splicing, and is classified under keywords including Disease variant and Dystonia, suggesting relevance to movement disorders.
- Required for neuronal maintenance, particularly in the central nervous system.
Experimental Guidance and Technical Tips
- The immunogen (aa 54-373 of human MECR) covers a large region, likely generating polyclonal antibodies recognizing multiple epitopes. For IHC-P, consider antigen retrieval protocols and tissue-specific fixation conditions. For ELISA, validate antibody pairing with a suitable detection system.
- Cross-reactivity is reported for human; reactivity with other species should be verified in the relevant sample system.
- The predicted molecular weight is 40 kDa (UniProt: 40462 Da); confirm by western blot if used.
