Biological Background: MECP2 Function and Localization
- Encodes Methyl-CpG-binding protein 2, a chromosomal protein essential for reading DNA methylation marks and regulating transcription.
- Binds specifically to single methylated CpG pairs, mediating transcriptional repression via recruitment of histone deacetylase and the SIN3A corepressor complex.
- Recognizes both 5-methylcytosine (5mC) and 5-hydroxymethylcytosine (5hmC) with higher affinity for 5mC, linking DNA methylation to gene silencing.
- Localizes predominantly to the nucleus, consistent with its role in chromatin interaction and epigenetic regulation.
- Ubiquitously expressed in adult somatic tissues, highlighting its fundamental function in cellular epigenetic maintenance.

Experimental Guidance and Technical Tips
- The immunogen was raised against a synthetic peptide spanning amino acids 387–486 in the C-terminal region of human MECP2. Cross-reactivity with mouse and rat is predicted due to sequence conservation, but validation in the target species is advised.
- For Western blotting, expect a band at approximately 52 kDa. Use nuclear-enriched lysates from human, mouse, or rat cells/tissues as positive controls and confirm specificity.
- In immunofluorescence/ICC experiments, MECP2 shows nuclear staining. Optimize permeabilization and fixation protocols for nuclear antigens to ensure accurate localization.
- For ELISA applications, the antibody can detect MECP2 peptides or recombinant protein. Standard direct or indirect ELISA protocols are suitable; titrate antibody concentration for optimal signal-to-noise ratio.
