Biological Background: MDM2 Function and Localization
- MDM2 (also known as Hdm2, HDMX) is an E3 ubiquitin-protein ligase that serves as a critical negative regulator of the tumor suppressor p53/TP53, mediating its ubiquitination and proteasomal degradation. Related references: PMID:29681526
- It inhibits p53/TP53- and p73/TP73-mediated cell cycle arrest and apoptosis by binding to their transcriptional activation domains, and promotes nuclear export of p53.
- MDM2 also acts as a ubiquitin ligase toward itself (auto-ubiquitination) and other substrates including ARRB1, RB1, DAXX, DYRK2, IGF1R, SNAI1, DCX, DLG4, and NDUFS1, regulating diverse cellular processes such as receptor endocytosis, mitochondrial respiration, and apoptosis. - The protein localizes predominantly to the nucleus, including the nucleoplasm and nucleolus, but can also be found in the cytoplasm, reflecting its dynamic roles in different compartments.

- The immunogen used is a synthetic peptide corresponding to an internal region (amino acids 231-330) of human MDM2. This design may enable recognition of multiple isoforms and processed forms of MDM2.
- For Western blotting (WB), a band at approximately 55 kDa is expected; consider validating specificity using positive and negative cell lysates.
- For immunohistochemistry (IHC-P) and ELISA, optimize antibody dilution and detection conditions, as staining patterns may vary with tissue type and fixation methods.
- Given the sequence homology, cross-reactivity with mouse MDM2 is anticipated; validate in relevant mouse model systems if needed.
