Biological Background: MDM2 Function and Localization

  • MDM2 (also known as Hdm2, HDMX) is an E3 ubiquitin-protein ligase that serves as a critical negative regulator of the tumor suppressor p53/TP53, mediating its ubiquitination and proteasomal degradation. Related references: PMID:29681526
  • It inhibits p53/TP53- and p73/TP73-mediated cell cycle arrest and apoptosis by binding to their transcriptional activation domains, and promotes nuclear export of p53.
  • MDM2 also acts as a ubiquitin ligase toward itself (auto-ubiquitination) and other substrates including ARRB1, RB1, DAXX, DYRK2, IGF1R, SNAI1, DCX, DLG4, and NDUFS1, regulating diverse cellular processes such as receptor endocytosis, mitochondrial respiration, and apoptosis.
