Biological Background: MDM2 Function and Localization
- MDM2 is an E3 ubiquitin-protein ligase that mediates ubiquitination of p53/TP53, leading to its proteasomal degradation. It inhibits p53- and p73-mediated cell cycle arrest and apoptosis by directly binding their transcriptional activation domains. Related references: PMID:29681526
- MDM2 also acts as an E3 ubiquitin ligase toward itself and ARRB1, permits the nuclear export of p53, promotes ubiquitin-independent degradation of RB1, and inhibits DAXX-mediated apoptosis via ubiquitination and degradation.

- The immunogen comprises amino acids 200–430 of human MDM2, covering the acidic domain, zinc finger, and RING finger regions conserved across human, mouse, and rat. For Western blotting, consider using lysates from MDM2-overexpressing cancer cell lines (e.g., SJSA-1) as positive controls, and validate detection in mouse and rat tissues with appropriate species-specific secondary antibodies.
- A primary band of ~55 kDa is expected in WB; higher molecular weight species may represent post-translationally modified forms (e.g., ubiquitinated or phosphorylated MDM2). Including phosphatase inhibitors during sample preparation may help preserve the expected molecular weight.
- For ELISA, test the antibody in both direct antigen coating and sandwich formats. Preliminary checkerboard titrations are recommended to optimize coating antigen and antibody working concentrations.
