Biological Background: MDM2 Function and Localization

  • MDM2, also known as Hdm2, is an E3 ubiquitin-protein ligase that catalyzes the ubiquitination of p53/TP53, targeting it for proteasomal degradation (PubMed:29681526). Related references: PMID:29681526

  • As an oncoprotein, MDM2 inhibits p53/TP53- and p73/TP73-mediated cell cycle arrest and apoptosis by binding their transcriptional activation domains.

  • The protein also ubiquitinates itself and ARRB1, and mediates proteasome-dependent degradation of RB1 protein, thereby modulating cell cycle progression.
