Biological Background: MDC1 Function and Localization
- MDC1 (Mediator of DNA damage checkpoint protein 1), also known as NFBD1 (Nuclear factor with BRCT domains 1), acts as a histone reader that specifically binds phosphorylated histone H2AX (γH2AX) at sites of DNA double-strand breaks, serving as a molecular scaffold for DNA repair and signal transduction proteins. - Upon DNA damage, MDC1 facilitates the recruitment and retention of DNA repair factors, including the MRN complex (via NBN) and TOPBP1, to damage foci; it is essential for the subsequent phosphorylation and activation of checkpoint kinases ATM, CHEK1, and CHEK2, leading to cell cycle arrest and stabilization of p53. - For Western blot (WB), due to the large molecular weight (~227 kDa), consider using low-percentage SDS-PAGE gels (e.g., 6-8% separating gel) and sufficient transfer time.
- For immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), validate staining patterns in known positive tissues (e.g., testis) or DNA-damage-induced cell models.
- The antibody has been reported for ELISA, but optimal working dilutions must be determined empirically by the end user.
