Biological Background: MCOLN2/TRPML2 Function and Localization
- MCOLN2 encodes mucolipin-2, also known as transient receptor potential channel mucolipin 2 (TRPML2), a member of the TRPML subfamily of ion channels.
- TRPML2 functions as a nonselective cation channel with inwardly rectifying activity, permeable to Ca2+ and Fe2+, and plays a role in the regulation of membrane trafficking events. Related references: PMID:19885840, PMID:19940139
- It may activate ARF6 and participate in the trafficking of GPI-anchored cargo proteins to the cell surface via the ARF6-regulated recycling pathway. Related references: PMID:17662026
- In the immune system, TRPML2 and TRPML1 may have redundant roles in specialized lysosomes of B cells in adaptive immunity, and in innate immunity, it may regulate chemokine secretion and macrophage migration (By similarity).
- TRPML2 can heteromerize with MCOLN1, potentially contributing to many lysosome-dependent cellular events in a tissue-specific manner. Related references: PMID:19885840
- Subcellular localization includes the cell membrane, late endosome membrane, lysosome membrane, and recycling endosome membrane, consistent with its roles in trafficking and ion homeostasis.
- Keywords from UniProt highlight its involvement in calcium transport, ion transport, protein transport, immunity, and lysosome biology.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 90-275 of human MCOLN2, located in the extracellular/luminal domain, potentially providing specificity for detection applications.
- For Western blot, the predicted molecular weight is 66 kDa; however, post-translational modifications may result in observed bands at different sizes. Include appropriate controls from human or rat samples.
- For ELISA, validate the antibody in the relevant sample matrix to confirm specificity and sensitivity.
- As a polyclonal antibody, it may recognize multiple epitopes and could be considered for applications requiring robust target detection.
