Biological Background: DNA replication licensing factor MCM6 Function and Localization
- DNA replication licensing factor MCM6 (p105MCM) is a core component of the MCM2-7 complex, which acts as the replicative helicase essential for 'once per cell cycle' DNA replication initiation and elongation in eukaryotic cells.
- MCM6, together with other MCM proteins, forms the CDC45-MCM-GINS (CMG) helicase, the molecular machine that unwinds template DNA during replication and serves as the foundation for replisome assembly (PubMed:16899510, PubMed:32453425, PubMed:34694004). Related references: PMID:16899510, PMID:32453425, PMID:34694004
- The active ATPase sites in the MCM2-7 ring are formed at subunit interfaces; a conserved arginine finger motif from one subunit is provided in trans to the ATP-binding site of the adjacent subunit, and these six ATPase sites contribute differentially to helicase activity (PubMed:32453425). Related references: PMID:32453425
- MCM6 is predominantly localized in the nucleus during G1 phase and associates with chromatin, consistent with its role in DNA replication licensing.
- The protein undergoes various post-translational modifications including acetylation and phosphorylation, which may regulate its activity during the cell cycle.
- According to UniProt, MCM6 is classified under keywords such as ATP-binding, Cell cycle, DNA replication, DNA-binding, Glycoprotein, Helicase, Hydrolase, Nucleotide-binding, Phosphoprotein, and Reference proteome.
Experimental Guidance and Technical Tips
- Since the immunogen corresponds to amino acids 601-821, the antibody recognizes a C-terminal region of human MCM6; consider validating detection of any relevant cleavage products or isoforms in your experimental system.
- For WB, a positive control such as HeLa or MCF-7 cell lysate (human) or NIH/3T3 lysate (mouse) is recommended; optimize antibody dilution and blocking conditions for your specific sample.
- The antibody has been used for knockout/knockdown validation (KD Validated), indicating reliable detection of endogenous MCM6; treat this as supporting specificity data.
- For ELISA, assess the antibody’s performance as a capture or detection reagent; pair with a suitable anti-MCM6 antibody or use recombinant MCM6 protein as standard.
- The expected band size is ~93 kDa; use appropriate gel percentage (e.g., 8% SDS-PAGE) and molecular weight markers for clear separation.
