Biological Background: MCM5 Function and Localization
-
MCM5 (CDC46/P1-CDC46) is the DNA replication licensing factor and a core component of the MCM2-7 complex, the eukaryotic replicative helicase essential for once-per-cell-cycle DNA replication initiation and elongation. Related references: PMID:40940420
-
Together with CDC45 and GINS, MCM5 forms the CDC45-MCM-GINS (CMG) helicase that unwinds duplex DNA at replication forks, serving as the central platform for replisome assembly. Related references: PMID:16899510, PMID:32453425, PMID:34694004
-
The MCM2-7 ring contains six ATPase active sites formed at subunit interfaces; MCM5 contributes a conserved arginine finger motif to the ATP-binding pocket of the neighboring subunit, with differential contributions to helicase activity. Related references: PMID:32453425
-
MCM5 localizes to the nucleus and is chromatin-bound in a cell cycle-dependent manner, playing a critical role in replication licensing and S-phase progression.
-
Post-translational modifications including acetylation, phosphorylation, and ubiquitin-like conjugation regulate MCM5 stability and activity.
-
MCM5 is essential for genome stability; mutations in the MCM5 gene are associated with Meier-Gorlin syndrome 8 (MGORS8) and other developmental disorders.
-
MCM5 is a member of the MCM family of ATP-dependent helicases, sharing conserved domains with MCM2-7, and is highly expressed in proliferating cells.
-
The protein has a predicted molecular weight of ~82 kDa and contains nucleotide-binding motifs critical for its enzymatic function.
Experimental Guidance and Technical Tips
- The immunogen (recombinant protein spanning amino acids 1–230 of human MCM5) targets the N-terminal region, which contains conserved motifs; cross-reactivity with mouse and rat orthologs has been observed. For species not listed, verify sequence homology before use.
- For Western blot (WB), consider a starting dilution of 1:500–1:2000 and use nuclear or whole-cell lysates from proliferating cells, where MCM5 is abundant. The predicted band is ~82 kDa; note that phosphorylation may cause slight mobility shifts.
- In immunofluorescence/immunocytochemistry (IF/ICC), MCM5 exhibits nuclear, cell cycle-dependent staining; optimize fixation and permeabilization conditions to preserve chromatin-bound protein.
- For immunoprecipitation (IP), the antibody can enrich native MCM5 and its associated MCM complex; validate co-IP of other MCM subunits to confirm complex integrity.
