Biological Background: MCM4 Function and Localization
- MCM4 is a core subunit of the MCM2-7 replicative helicase complex and the CDC45-MCM-GINS (CMG) helicase, driving DNA unwinding during replication. Related references: PMID:16899510, PMID:25661590, PMID:32453425
- The MCM2-7 ring’s ATPase activity is generated by composite active sites at subunit interfaces, where a conserved arginine finger motif is provided by the adjacent subunit. Related references: PMID:16899510, PMID:25661590, PMID:32453425
- MCM4 localizes to the nucleus and chromosomes, consistent with its role in genome duplication.
- Post-translational modifications including acetylation, phosphorylation, and ubiquitination may regulate MCM4 activity.
- MCM4 is also known as CDC21 homolog and P1-CDC21, reflecting its evolutionary conservation.
Experimental Guidance and Technical Tips
- The immunogen corresponds to a C-terminal peptide (amino acids 764–863) of human MCM4; this region may facilitate recognition of full-length MCM4 across species.
- For Western blot, the expected band is around 97 kDa; consider using a resolving gel appropriate for high molecular weight proteins.
- In ELISA, titration against the immunizing peptide or recombinant MCM4 is recommended to optimize signal-to-noise ratio.
- Cross-reactivity is reported for human, mouse, and rat; performing sequence alignment for other species is advisable before use.
