Biological Background: MCM3 Function and Localization

  • MCM3 is a DNA replication licensing factor, also known as DNA polymerase alpha holoenzyme-associated protein P1, P1-MCM3, RLF subunit beta, or p102.
  • It is a core component of the MCM2-7 complex, which functions as the replicative helicase essential for once-per-cell-cycle DNA replication initiation and elongation.
  • MCM3 is a subunit of the CDC45-MCM-GINS (CMG) helicase, the molecular machine that unwinds template DNA during replication and around which the replisome is built. Related references: PMID:32453425 PMID:34694004 PMID:34700328
  • The active ATPase sites in the MCM2-7 ring are formed by interaction surfaces of neighboring subunits, with a conserved arginine finger motif provided in trans to the ATP-binding site of the adjacent subunit.   • MCM3 is required for entry into S phase and for cell division.
