Biological Background: MCM3 Function and Localization
- MCM3 is a DNA replication licensing factor, also known as DNA polymerase alpha holoenzyme-associated protein P1, P1-MCM3, RLF subunit beta, or p102.
- It is a core component of the MCM2-7 complex, which functions as the replicative helicase essential for once-per-cell-cycle DNA replication initiation and elongation.
- MCM3 is a subunit of the CDC45-MCM-GINS (CMG) helicase, the molecular machine that unwinds template DNA during replication and around which the replisome is built. Related references: PMID:32453425 PMID:34694004 PMID:34700328
- The active ATPase sites in the MCM2-7 ring are formed by interaction surfaces of neighboring subunits, with a conserved arginine finger motif provided in trans to the ATP-binding site of the adjacent subunit. The six ATPase active sites contribute differentially to helicase activity. Related references: PMID:32453425
- MCM3 is required for entry into S phase and for cell division.
- Subcellular localization is primarily nuclear, associated with chromosomes.
- The protein is subject to acetylation, alternative splicing, phosphorylation, and glycosylation, as indicated by UniProt keywords.
- It has a molecular weight of approximately 91 kDa (calculated 90,981 Da from UniProt).
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 1-100 of human MCM3. For western blot, expect a band around 91 kDa. Validate in the relevant sample system.
- For IHC-P and IF/ICC, consider antigen retrieval and validation in appropriate tissue samples.
- For IP and ELISA, use standard protocols and confirm specificity in human, mouse, or rat samples.
