Biological Background: MCM2 Function and Localization

  • DNA replication licensing factor MCM2 (minichromosome maintenance protein 2 homolog; also known as BM28, CCNL1, CDCL1) is a core component of the MCM2-7 complex, which is the putative replicative helicase.
  • The MCM2-7 complex forms the core of the CDC45-MCM-GINS (CMG) helicase, the molecular machine responsible for unwinding template DNA during replication, and functions in both initiation and elongation phases (PubMed:32453425, PubMed:34694004, PubMed:34700328). Related references: PMID:32453425 PMID:34694004 PMID:34700328
  • Active ATPase sites in the MCM2-7 ring are formed at the interface of neighboring subunits, with a conserved arginine finger motif provided in trans relative to the ATP-binding site of the adjacent subunit (PubMed:32453425). Related references: PMID:32453425
  • MCM2 is required for entry into S phase and for cell division (PubMed:8175912). Related references: PMID:8175912
  • Plays a role in the development of terminally differentiated hair cells in the cochlea and induces cell apoptosis (PubMed:26196677). Related references: PMID:26196677
  • Localizes to the nucleus and associates with chromosomes during DNA replication.
  • Post-translational modifications include phosphorylation, acetylation, and ubiquitination; the protein contains zinc-finger motifs and exhibits ATP-binding and DNA-binding activities.
  • Mutations in MCM2 are associated with autosomal dominant non-syndromic hearing loss (DFNA70).

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the N-terminal region (amino acids 1–100) of human MCM2. Consider using the recommended peptide blocking strategy for specificity control in relevant assays.
  • For Western blot (WB), expect a band at approximately 102 kDa. Optimize reducing/denaturing conditions and use appropriate loading controls.
  • For immunohistochemistry (IHC-P), antigen retrieval methods (e.g., heat-induced epitope retrieval) may be required to expose the epitope; validate with known positive and negative tissue controls.
  • For immunofluorescence/immunocytochemistry (IF/ICC), fixation and permeabilization conditions should be empirically determined, as the nuclear localization of MCM2 may require optimized access.
  • For ELISA, coating with the immunogenic peptide may serve as a positive control; confirm binding under your specific assay conditions.
  • Although cross-reactivity with mouse and rat is reported, we recommend verifying specificity in your experimental system, as epitope conservation may vary.
