Biological Background: MC1 Receptor Function and Localization

  • MC1R is a G protein-coupled receptor that binds melanocyte-stimulating hormones (alpha, beta, gamma-MSH) and adrenocorticotropic hormone (ACTH), peptide products of the POMC precursor. Related references: PMID:11442765, PMID:11707265, PMID:1325670
  • Ligand binding activates the G(s) protein, stimulating adenylate cyclase and the cAMP-dependent pathway, thereby promoting melanogenesis (production of eumelanin and pheomelanin). Related references: PMID:11707265, PMID:1325670, PMID:16463023
  • Interaction with G protein-coupled receptor opsin 3 (OPN3) couples to G(i) proteins and inhibits the alpha-MSH-induced cAMP response, reducing melanin synthesis. Related references: PMID:31097585
