Biological Background: MC1 Receptor Function and Localization
- MC1R is a G protein-coupled receptor that binds melanocyte-stimulating hormones (alpha, beta, gamma-MSH) and adrenocorticotropic hormone (ACTH), peptide products of the POMC precursor. Related references: PMID:11442765, PMID:11707265, PMID:1325670
- Ligand binding activates the G(s) protein, stimulating adenylate cyclase and the cAMP-dependent pathway, thereby promoting melanogenesis (production of eumelanin and pheomelanin). Related references: PMID:11707265, PMID:1325670, PMID:16463023
- Interaction with G protein-coupled receptor opsin 3 (OPN3) couples to G(i) proteins and inhibits the alpha-MSH-induced cAMP response, reducing melanin synthesis. Related references: PMID:31097585
- Binding of Agouti/ASP precludes alpha-MSH-induced signaling, downregulating melanogenesis (By similarity).
- Interaction with MGRN1 displaces G(s) protein, further suppressing MC1R signaling. Related references: PMID:19737927
- Subcellular localization: cell membrane.
- Expressed in melanocytes and corticoadrenal tissue. Related references: PMID:1325670, PMID:31097585
- Contains structural features: disulfide bonds, palmitoylation sites, and glycosylation.
Experimental Guidance and Technical Tips
- The immunogen (amino acids 200-317) targets an intracellular C-terminal region of human MC1R; consider this when designing experiments for detecting endogenous receptor.
- For WB, the predicted molecular weight is 35 kDa; validate in relevant sample systems.
- For ELISA, the antibody may be suitable for detecting MC1R peptides; serial dilutions are recommended for optimal signal.
- As a polyclonal antibody, it may recognize multiple epitopes; cross-reactivity with mouse and rat is reported, enabling multi-species studies.
