Biological Background: MBNL1 Function and Localization
- Protein identity and isoforms: Muscleblind-like protein 1 (MBNL1), also known as Triplet-expansion RNA-binding protein, is encoded by the MBNL1 gene (EXP, KIAA0428, MBNL). Multiple isoforms are produced via alternative splicing.
- Core function in pre-mRNA splicing: MBNL1 mediates pre-mRNA alternative splicing, acting as both activator and repressor. It inhibits cardiac troponin-T (TNNT2) exon inclusion while promoting insulin receptor (IR) exon inclusion in muscle, and antagonizes CELF protein splicing activities.
- Splicing regulatory mechanism: MBNL1 binds the 5'-YGCU(U/G)Y-3' consensus sequence and inhibits spliceosome A complex formation on TNNT2 intron 4. It also binds expanded CUG repeat RNA, relevant to myotonic dystrophy.
- Subcellular localization: Localizes to the nucleus, cytoplasm, and cytoplasmic granules.
- Tissue-specific expression: Highly expressed in cardiac and skeletal muscle and during myoblast differentiation; weakly expressed in brain, placenta, lung, liver, kidney, and pancreas.
- Pathophysiological roles: Regulates NCOR2 alternative splicing and, together with RBPMS and RBFOX2, activates alternative splicing in vascular smooth muscle cells.

Experimental Guidance and Technical Tips
- Given the immunogen sequence (aa 289-388), the antibody may recognize an epitope in the C-terminal region of MBNL1; ensure the target of interest contains this region, especially when working with specific isoforms.
- For western blot (WB) detection, a band around 42 kDa is expected; human heart or skeletal muscle lysates may serve as positive controls due to high MBNL1 expression. Validate in the relevant sample system.
- In immunofluorescence (IF/ICC), nucleocytoplasmic localization with possible granular staining is anticipated; fixation and permeabilization conditions should be optimized for the cell type under study.
- Since cross-reactivity is reported only for human, species reactivity should be independently verified if used with other species' samples.
