  • Tryptase alpha/beta-1 (UniProt: Q15661), also known as tryptase I or tryptase alpha-1, is a serine protease encoded by the TPSAB1 gene. It shares high sequence homology with TPSB2 (tryptase beta-2).
  • It functions as the major neutral protease in mast cell secretory granules and is released during mast cell degranulation, playing a key role in inflammatory and allergic responses.
  • The protease is implicated in innate immunity through the cleavage of various substrates. Isoform 2 (also referred to as tryptase beta-1) exhibits differential substrate specificity, more efficiently cleaving large substrates such as fibronectin but less efficiently processing small substrates compared to isoform 1.

  • In immunohistochemistry (IHC-P), robust staining is expected in mast cell-rich tissues such as skin, lung, and gastrointestinal mucosa. Antigen retrieval methods (e.g., heat-induced epitope retrieval) are recommended, and appropriate isotype controls should be included.
