Biological Background: MASP1 Function and Localization
- MASP1 is the precursor of a serum protease that activates the lectin pathway of the complement system. Following binding of lectins (e.g., MBL2, ficolins) to carbohydrates on pathogen surfaces, MASP1 undergoes autoproteolytic processing and then cleaves and activates MASP2, the next component in the cascade. - MASP1 also plays a role in the alternative complement pathway. Specifically, isoform MASP-3 catalyzes cleavage and activation of factor D (CFD), the initiating protease of the alternative pathway, which serves as an amplification loop enhancing other complement pathways. - The protein is primarily expressed by the liver and secreted into the plasma. It can also be found on cell surfaces.
- MASP1 gene encodes multiple isoforms through alternative splicing, including MASP-1, MASP-3, and MAp44, which have distinct functions in complement regulation and coagulation.
- It is a calcium-dependent serine protease featuring CUB, EGF-like, CCP (Sushi), and serine protease domains. It undergoes autocatalytic cleavage and is secreted as a glycoprotein. (Keywords: complement activation, innate immunity, disease variant.)
Experimental Guidance and Technical Tips
- The immunogen peptide maps to the N-terminal region (aa 20-300) of human MASP1, a sequence conserved in human, mouse, and rat, consistent with the reported cross-reactivity.
- For Western blot, consider using reduced-denaturing conditions; the full-length protein has a predicted molecular weight of ~79 kDa, but additional bands may appear due to isoforms (MASP-3 ~80 kDa, MAp44 ~44 kDa) or cleavage products.
- For ELISA, validate the antibody in the relevant sample matrix (e.g., serum or plasma) to confirm sensitivity and specificity for native MASP1.
- As a polyclonal antibody, batch-to-batch consistency should be verified, and appropriate controls (e.g., knockout samples or blocking peptides) are recommended for assay validation.
