Biological Background: MARK2 Function and Localization
-
MARK2 is a serine/threonine-protein kinase also known as EMK1, PAR-1, and Par-1b, encoded by the MARK2 gene in humans. It belongs to the MARK family of kinases.
-
It plays a key role in regulating cell polarity by phosphorylating microtubule-associated proteins (MAPs) such as MAP2, MAP4, and MAPT/TAU at KXGS motifs, leading to their detachment from microtubules and subsequent disassembly.

-
MARK2 phosphorylates various downstream targets including CRTC2/TORC2, DCX, HDAC7, KIF13B, and RAB11FIP2, thereby regulating processes like neuronal migration, axogenesis, histone deacetylase localization, and epithelial cell polarity.
-
In the nervous system, MARK2 is critical for neurite outgrowth and the establishment of neuronal polarity. It modulates neuronal migration by phosphorylating DCX and controls axonal transport via KIF13B phosphorylation.
-
Subcellularly, MARK2 localizes to the cell membrane (particularly lateral cell membrane), cytoplasm, cytoskeleton, and dendrites. Its localization reflects its roles in membrane trafficking and cytoskeletal organization.
-
MARK2 positively regulates the Wnt signaling pathway, likely by phosphorylating disheveled proteins (DVL1, DVL2, DVL3), thereby modulating developmental decisions.
-
It is involved in autophagy regulation: during ER stress or starvation, MARK2 phosphorylates UBAC2 at Ser-223, promoting UBAC2 dimerization and interaction with GABARAP to mediate ER-phagy.

-
MARK2 is highly expressed in heart, brain, skeletal muscle, and pancreas, with lower expression in lung, liver, and kidney. It is associated with diseases such as autism spectrum disorder and intellectual disability, underscoring its importance in neural function.
Experimental Guidance and Technical Tips
- The immunogen corresponds to a region (amino acids 350-455) within the kinase domain of MARK2, which is conserved across human, mouse, and rat, supporting the observed cross-reactivity.
- For Western blot (WB), consider that the predicted molecular weight is 88 kDa, but actual migration may vary due to phosphorylation or splice variants. Validate using appropriate positive controls (e.g., brain or heart tissue lysates).
- In immunofluorescence (IF/ICC), MARK2 can be detected at the cell membrane, cytoplasm, and in cell projections like dendrites, so fixation and permeabilization conditions should be optimized to preserve these structures.
- For ELISA applications, pair this antibody with appropriate secondary detection reagents and validate using recombinant MARK2 protein or cell lysates expressing the target.
