Biological Background: MARCO Function and Localization
- Macrophage receptor MARCO (also known as SR-A6, SCARA2) is a member of the scavenger receptor class A family characterized by a collagenous structure.
- Functions as a pattern recognition receptor (PRR) that binds both Gram-positive and Gram-negative bacteria, playing a key role in innate immunity (PubMed:9468508). Related references: PMID:9468508
- Binds the secretoglobin SCGB3A2, which may modulate immune responses (PubMed:12847263). Related references: PMID:12847263
- Also involved in binding of unopsonized particles by alveolar macrophages (By similarity).
- Localizes to the cell membrane as a transmembrane receptor.
- Expressed predominantly in tissue macrophages, including alveolar macrophages, Kupffer cells of the liver, and macrophages in thymus, kidney, and spleen (PubMed:9468508).
- Contains a collagen domain, disulfide bonds, and N-glycosylation sites, and exists in alternatively spliced forms.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 421-520 of human MARCO, located in the C-terminal extracellular region; the resulting polyclonal antibody likely recognizes multiple epitopes.
- For Western blotting, the predicted protein weight is ~53 kDa, but the observed band may shift due to post-translational modifications such as glycosylation; optimization of reducing/non-reducing conditions may be necessary.
- For immunofluorescence (IF-P), appropriate fixation and permeabilization protocols should be tested, as MARCO is a transmembrane protein.
- For ELISA, validate antibody binding using recombinant MARCO protein or cell lysates from macrophage cell lines.
- Cross-reactivity with mouse and rat has been reported, but species-specific validation in the experimental system is recommended.
