Biological Background: MARCKSL1 Function and Localization
- MARCKSL1 (MARCKS-like protein 1), also named MacMARCKS, MLP, or MRP, is encoded by the MARCKSL1 gene in humans. It is related to the MARCKS family of actin-binding proteins involved in cell motility.
- The protein regulates cell movement by controlling actin cytoskeleton homeostasis, filopodium and lamellipodium formation. Unphosphorylated MARCKSL1 promotes cell migration, while phosphorylation by MAPK8 (JNK1) induces actin bundle formation and stabilization, reducing actin plasticity and restricting movement, including neuronal migration. - MARCKSL1 is implicated in coupling protein kinase C and calmodulin signal transduction systems, acting as a substrate in phosphorylation-dependent signaling cascades.
- It localizes to the cytoplasm, associates with the cytoskeleton, and is found at the cell membrane, reflecting its role in linking membrane dynamics to actin reorganization.
- Post-translational modifications include N-terminal myristoylation, which aids membrane targeting, and phosphorylation on multiple residues that modulate its activity.

- The immunogen is a synthetic peptide spanning amino acids 95–195, which may not encompass the N-terminal myristoylation site; consider this when detecting full-length or modified protein.
- For Western blot, the expected molecular weight is ~19 kDa, but shifts can occur due to phosphorylation or other modifications. Validate band specificity with appropriate controls.
- In ELISA applications, confirm that the peptide immunogen is compatible with your plate-coating and detection conditions.
