Biological Background: MARCKS Function and Localization
- MARCKS (Myristoylated alanine-rich C-kinase substrate) is a membrane-associated protein that plays a key role in modulating the actin cytoskeleton, regulating processes such as chemotaxis, cell motility, adhesion, phagocytosis, and exocytosis. Related references: PMID:23704996, PMID:36009319
- Acts as a phosphatidylinositol 4,5-bisphosphate (PIP2) sequestering protein at lipid rafts in the plasma membrane; this sequestration is reversed by protein kinase C, leading to exocytosis inhibition. Related references: PMID:23704996
- During inflammation, MARCKS promotes migration and adhesion of inflammatory cells and enhances secretion of cytokines such as TNF, particularly in macrophages. Related references: PMID:37949888
- Essential for bacteria-induced intracellular reactive oxygen species (ROS) formation in monocytic cells.
- Contributes to neurite initiation and outgrowth by interacting with CDC42, a regulator of cell shape and cytoskeletal dynamics (By similarity).
