Biological Background: MAPT Function and Localization

  • MAPT (Microtubule-associated protein tau), also known as Tau or PHF-tau, is a neuronal microtubule-associated protein encoded by the Mapt gene in mouse (Gene ID: 17762) and MAPT in human (Gene ID: 4137).
  • Tau promotes microtubule assembly and stability, and is thought to be involved in establishing and maintaining neuronal polarity. It binds axonal microtubules via its C-terminus and neural plasma membrane components via its N-terminus, suggesting a linker function.
  • Multiple isoforms are generated by alternative splicing. Shorter isoforms impart cytoskeletal plasticity while longer isoforms promote stabilization. Isoform PNS-tau is expressed in the peripheral nervous system, with others in the CNS.
  • Subcellular localization includes cytoplasm, cytosol, cell membrane, cytoskeleton, axonal and dendritic projections, and it can also be secreted.

  • The immunogen corresponds to the N-terminal region (amino acids 3-214) of mouse MAPT, which may generate antibodies recognizing multiple isoforms spanning the predicted molecular weights of ~35-38 kDa, 44 kDa, and 76 kDa. When performing Western blotting, expect bands within these ranges depending on the isoform expressed in your sample.
  • For immunofluorescence/immunocytochemistry (IF/ICC), consider optimizing fixation and permeabilization conditions, as tau localizes to multiple subcellular structures including microtubules, cytoplasmic compartments, and the plasma membrane. Co-staining with a neuronal marker may help verify specificity.
  • As a polyclonal antibody, batch-to-batch consistency should be validated in the relevant experimental system. Users are advised to titrate the antibody concentration for ELISA to achieve optimal signal-to-noise ratios.
