Biological Background: MAPKAPK-2/MK2 Function and Localization

  • MAPKAPK-2/MK2 is a stress-activated serine/threonine-protein kinase involved in multiple cellular processes, including cytokine production, cytoskeleton reorganization, cell migration, cell cycle control, chromatin remodeling, DNA damage response, and transcriptional regulation.
  • Upon stress, it is phosphorylated and activated by p38-alpha/MAPK14, leading to phosphorylation of downstream substrates on serine residues within the consensus sequence Hyd-X-R-X(2)-S.
  • It phosphorylates HSF1, promoting interaction with HSP90 and inhibiting HSF1 homotrimerization, DNA-binding, and transactivation activities, thereby modulating the heat shock response. Related references: PMID:16278218
  • In response to UV irradiation, it phosphorylates CEP131, which promotes binding of CEP131 to 14-3-3 proteins and inhibits formation of novel centriolar satellites. Related references: PMID:26616734
