Biological Background: MAPK8 Function and Localization
- MAPK8 (JNK1/SAPK1c) is a stress-activated serine/threonine kinase that is central to the SAP/JNK signaling cascade, responding to cytokines and environmental stress. Related references: PMID:28943315
- Upon activation by dual phosphorylation from MAP2K4/MKK4 and MAP2K7/MKK7, MAPK8 phosphorylates transcription factors JUN, JDP2, and ATF2 to regulate AP-1 transcriptional activity. Related references: PMID:18307971
- In the nucleus, MAPK8 phosphorylates the replication licensing factor CDT1, disrupting its interaction with HBO1 and inhibiting replication origin acetylation, thus controlling DNA replication initiation. Related references: PMID:21856198
