Biological Background: MAPK6 Function and Localization

  • MAPK6 (also known as extracellular signal-regulated kinase 3, ERK-3, or p97-MAPK) is an atypical mitogen-activated protein kinase encoded by the MAPK6 gene.
  • It functions in a non-classical MAP kinase cascade, phosphorylating microtubule-associated protein 2 (MAP2) and MAPKAPK5.
  • The formation of a complex with MAPKAPK5 leads to reciprocal phosphorylation: MAPK6 is phosphorylated at Ser-189, mediating the activation of MAPKAPK5, which in turn phosphorylates MAPK6. This signaling module may promote cell cycle entry.
  • Subcellularly, MAPK6 is found in both the cytoplasm and nucleus.
  • Highest expression levels are observed in skeletal muscle and brain, with additional expression in heart, placenta, lung, liver, pancreas, kidney, and skin fibroblasts.
  • Key structural features include an ATP-binding domain and a serine/threonine kinase domain; post-translational modifications involve phosphorylation and ubiquitination.

Experimental Guidance and Technical Tips

  • The immunogen spans the C-terminal region (aa 372–721) of human MAPK6. For western blotting, the predicted molecular weight is ~83 kDa, but migration may be influenced by phosphorylation status; consider using appropriate phosphatase inhibitors during sample preparation.
  • This antibody has been reported to react with human, mouse, and rat MAPK6. It is advisable to validate specificity in the relevant species and tissue context before use.
  • For immunofluorescence/ICC, optimize fixation and permeabilization protocols to preserve both cytoplasmic and nuclear localization of MAPK6.
  • As a polyclonal antibody, it may recognize multiple epitopes, potentially including both phosphorylated and unphosphorylated forms; confirm band patterns with appropriate controls.
