- MAPK6 (also known as extracellular signal-regulated kinase 3, ERK-3, or p97-MAPK) is an atypical mitogen-activated protein kinase encoded by the MAPK6 gene.
- It functions in a non-classical MAP kinase cascade, phosphorylating microtubule-associated protein 2 (MAP2) and MAPKAPK5.
- The formation of a complex with MAPKAPK5 leads to reciprocal phosphorylation: MAPK6 is phosphorylated at Ser-189, mediating the activation of MAPKAPK5, which in turn phosphorylates MAPK6. This signaling module may promote cell cycle entry.
- Subcellularly, MAPK6 is found in both the cytoplasm and nucleus.
- Highest expression levels are observed in skeletal muscle and brain, with additional expression in heart, placenta, lung, liver, pancreas, kidney, and skin fibroblasts.
- Key structural features include an ATP-binding domain and a serine/threonine kinase domain; post-translational modifications involve phosphorylation and ubiquitination.
