Biological Background: MAPK1/MAPK3 Function and Localization
- MAPK1 (ERK2, p42-MAPK) and MAPK3 (ERK1, p44-MAPK) are the two prototypic mitogen-activated protein kinases (MAPKs) that constitute the core of the conserved MAPK/ERK signaling cascade.
- Upon activation, MAPK1/3 phosphorylate a broad range of nuclear and cytoplasmic substrates—including transcription factors (e.g., ELK1, FOS), cytoskeletal proteins, and other kinases—to regulate cell growth, proliferation, differentiation, adhesion, and survival.
- MAPK1 specifically phosphorylates GLI1 in response to smoothened signaling, promoting GLI1 dissociation from SUFU and its nuclear translocation, thereby positively regulating the Hedgehog pathway. Related references: PMID:35831023
- Under hypoxic conditions, MAPK1 phosphorylates phosphoglycerate kinase PGK1, directing it to mitochondria and suppressing acetyl-coenzyme A production from pyruvate. Related references: PMID:26942675

Experimental Guidance and Technical Tips
- This rabbit polyclonal antibody was raised against a synthetic peptide corresponding to amino acids 200-300 of human MAPK1 and MAPK3 (ERK2 and ERK1); it is predicted to recognize both kinases and may detect multiple bands in western blot due to isoforms and phospho-forms.
- For western blotting, load 20-30 µg of total protein lysate and use appropriate controls (e.g., serum-starved vs. growth factor-stimulated cells) to observe ERK activation. Expected molecular weights range from 36 to 43 kDa.
- In immunofluorescence/ICC, fix cells with 4% paraformaldehyde and permeabilize with 0.1% Triton X-100; primary antibody incubation overnight at 4°C is recommended. Validate staining specificity with peptide blocking or siRNA knockdown.
- For ELISA, this polyclonal antibody can be used as a capture or detection reagent; optimal coating and detection conditions should be determined empirically. A matched monoclonal antibody pair may improve sensitivity and specificity.
