Biological Background: MAP4K3 Function and Localization
- MAP4K3 (Mitogen-activated protein kinase kinase kinase kinase 3) is a serine/threonine kinase, also known as germinal center kinase-related protein kinase (GLK), MEK kinase kinase 3 (MEKKK 3), MAPKKKK3, and RAB8IPL1.
- It responds to environmental stress and acts upstream of the JUN N-terminal (JNK) signaling pathway. Related references: PMID:9275185
- Functions as an activator of the Hippo signaling pathway; it is partially redundant with STK3/MST2 and STK4/MST1 in phosphorylating LATS1/2, contributing to organ size control and tumor suppression. Related references: PMID:26437443
- Ubiquitously expressed, with highest levels in heart, brain, placenta, skeletal muscle, kidney, and pancreas; lower in lung and liver.
- Contains conserved kinase domains and is regulated by acetylation and phosphorylation events, as indicated by keywords ATP-binding, phosphoprotein, and acetylation.
- Alternative splicing generates multiple isoforms, potentially diversifying its functional roles.
- The immunogen maps to amino acids 755–894, lying within the C-terminal region of the protein, which may contribute to isoform specificity.
Experimental Guidance and Technical Tips
- The antibody is a polyclonal reagent raised against a recombinant C-terminal fragment (aa 755–894); consider verifying specificity via knockdown or blocking peptide experiments in your sample system.
- For Western blotting, the predicted molecular weight is approximately 101 kDa; use a gel percentage and transfer conditions suitable for high-molecular-weight proteins and include a pre-stained marker for reference.
- ELISA protocols may require optimization of antibody dilution and blocking; titrate the antibody using a known positive control.
- Cross-reactivity is demonstrated for human, mouse, and rat; validate in the specific tissue or cell type of interest to confirm species-specific performance.
- Due to the polyclonal nature, lot-to-lot variation can occur; it is advisable to reserve a reference lot for long-term studies.
