Biological Background: MAP4K2 Function and Localization
- MAP4K2 (also known as GCK, RAB8IP, BL44) is a serine/threonine-protein kinase that acts as a key MAP4K in the JNK and p38 MAPK signaling cascades.
- It is essential for innate immunity, being required for efficient JNK activation by TRAF6-dependent stimuli such as LPS, peptidoglycan, poly(I:C), and bacterial flagellin, and to a lesser extent by IL-1 and CD40 engagement.
- MAP4K2 functions in the Hippo signaling pathway by phosphorylating and activating LATS1/2, contributing to organ size control and tumor suppression. Related references: PMID:26437443
- The protein localizes to the cytoplasm, basolateral cell membrane, and Golgi apparatus membrane.
- Highly expressed in germinal center B-cells, with additional expression in lung, brain, and placenta.
- Involved in stress response, innate immunity, and possibly vesicle targeting or fusion; post-translational modifications include phosphorylation and ubiquitination.
