Biological Background: MAP3K14 Function and Localization
- MAP3K14, also known as NF-kappa-beta-inducing kinase (NIK) or HsNIK, is a serine/threonine-protein kinase encoded by the MAP3K14 gene.
- It functions as a lymphotoxin beta-activated kinase specifically involved in the non-canonical NF-kappa-B signaling pathway.
- Upon activation, MAP3K14 phosphorylates CHUK/IKKA, promoting proteolytic processing of NFKB2/p100 to p52, which leads to NF-kappa-B transcriptional activity. Related references: PMID:25406581 PMID:29230214
- This kinase plays an essential role in regulating genes for B-cell survival, lymphoid organogenesis, and immune responses. Related references: PMID:25406581
- MAP3K14 localizes to the cytoplasm and is weakly expressed in tissues such as testis, small intestine, spleen, thymus, peripheral blood leukocytes, prostate, ovary, and colon.
- Key biological properties include ATP binding, phosphorylation, and ubiquitin conjugation, and mutations in MAP3K14 are associated with disease variants.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 330-662 of human MAP3K14, a region within the kinase domain, suggesting specificity for the active or full-length protein.
- For Western blot, the predicted molecular weight is approximately 104 kDa; an 8-10% SDS-PAGE gel may be suitable for resolution.
- Cross-reactivity with rat is reported, so the antibody may be useful for studies in rodent models, but validation in the specific experimental context is recommended.
- Given the weak tissue expression, signal amplification strategies or enrichment steps might be considered for detection in tissue lysates.
