Biological Background: MAP3K1 Function and Localization
- MAP3K1, also known as MEKK1, MEK kinase 1, MAPKKK1, and aliases including SRXY6, is a serine/threonine-protein kinase that functions as a MAP kinase kinase kinase.
- It is a component of protein kinase signal transduction cascades, activating both the ERK and JNK pathways by phosphorylating MAP2K1 and MAP2K4, respectively. Related references: PMID:9808624
- MAP3K1 may also directly phosphorylate MAPK8/JNK1 kinase. Related references: PMID:17761173
- It plays a critical role in the NF-kappa-B signaling pathway by activating CHUK and IKBKB. Related references: PMID:9808624
- The protein contains an ATP-binding site, zinc-finger domain, and requires magnesium for kinase activity.
- Post-translational modifications include acetylation and phosphorylation; it is a phosphoprotein involved in multiple signaling networks.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide mapping to amino acids 1400-1512 of human MAP3K1 (NP_005912.1), which lies within the C-terminal kinase domain. Consider this when assessing isoform specificity.
- In Western blot, a band corresponding to the predicted molecular weight of ~164 kDa should be observed; ensure extended transfer times for high-molecular-weight proteins.
- For IHC-P applications, validate in appropriate human, mouse, or rat tissue samples, and optimize antigen retrieval protocols.
- This polyclonal antibody may detect multiple isoforms or cleavage products; verify banding patterns in relevant positive and negative controls.
