Biological Background: MAP2K4 Function and Localization

  • Dual specificity protein kinase that acts as an essential component of the stress-activated protein kinase/c-Jun N-terminal kinase (SAP/JNK) signaling pathway.
  • Together with MAP2K7/MKK7, is one of the only known kinases to directly activate the stress-activated protein kinases JNK1, JNK2, and JNK3 by phosphorylation.
  • Shows preference for phosphorylation of the tyrosine residue in the Thr-Pro-Tyr motif of JNKs, while MAP2K7 preferentially phosphorylates the threonine residue.
  • In addition to JNKs, activates the p38 MAPKs MAPK11, MAPK12, MAPK13, and MAPK14, broadening its role in cellular stress responses.
  • Required for maintaining peripheral lymphoid homeostasis; the MKK/JNK signaling pathway is involved in mitochondrial death signaling, including cytochrome c release and apoptosis.
  • Localizes to both the cytoplasm and nucleus.
  • Widely expressed with highest abundance in skeletal muscle.
  • Post-translational modifications include acetylation, phosphorylation, and methylation, which regulate its activity.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 300-399 of human MAP2K4; this region may confer cross-reactivity with mouse and rat orthologs, as indicated.
  • For Western blotting, a band of approximately 44 kDa is expected. Validate specificity using knockout (KO)-validated lysates or appropriate positive/negative controls.
  • For immunohistochemistry (paraffin), optimization of antigen retrieval and primary antibody dilution is recommended, considering tissue-specific expression patterns.
  • Although validated for human, mouse, and rat, cross-reactivity with other species should be tested empirically if desired.
