Biological Background: MAP1LC3A Function and Localization
- MAP1LC3A is a ubiquitin-like modifier essential for macroautophagy, participating in the elongation of the phagophore membrane during autophagosome formation.

- For Western blot (WB), note that endogenous MAP1LC3A can be detected as an unprocessed ~14 kDa form and a lipidated ~16 kDa form; consider optimizing lysis buffer, protease inhibitors, and gel conditions to resolve both species.
- When performing immunohistochemistry (IHC-P), validate fixation protocols and antigen retrieval methods for your tissue type; positive controls should include autophagic-rich tissues such as heart, brain, or liver.
- For ELISA applications, titrate the antibody to determine the optimal working concentration for either capture or detection setups, and evaluate cross-reactivity with closely related LC3 family members if needed.
