Biological Background: MAP1LC3A Function and Localization

  • MAP1LC3A is a ubiquitin-like modifier essential for macroautophagy, participating in the elongation of the phagophore membrane during autophagosome formation.
    Related references: PMID:20713600, PMID:24290141
  • Through its interaction with the reticulophagy receptor TEX264, MAP1LC3A drives the remodeling of endoplasmic reticulum subdomains into autophagosomes under nutrient stress, facilitating ER turnover via lysosomal degradation.
    Related references: PMID:31006537, PMID:31006538
  • It belongs to the ATG8 family and is also known as autophagy-related protein LC3 A, MAP1 light chain 3-like protein 1, or microtubule-associated proteins 1A/1B light chain 3A (MAP1A/MAP1B LC3 A).
  • The protein localizes mainly to cytoplasmic vesicles, autophagosome membranes, the endomembrane system, and the cytoskeleton, consistent with its role in vesicle trafficking and autophagy.
  • MAP1LC3A is post-translationally processed to expose a C-terminal glycine for conjugation with phosphatidylethanolamine (lipidation), and it is regulated by phosphorylation, reflecting its classification as a lipoprotein and phosphoprotein.
  • Tissue expression is highest in heart, brain, liver, skeletal muscle, and testis, while it is absent in thymus and peripheral blood leukocytes.
  • Alternative splicing generates multiple isoforms, and the protein is part of the ubiquitin-like conjugation pathway (Ubl conjugation) critical for autophagosomal membrane dynamics.

Experimental Guidance and Technical Tips

  • For Western blot (WB), note that endogenous MAP1LC3A can be detected as an unprocessed ~14 kDa form and a lipidated ~16 kDa form; consider optimizing lysis buffer, protease inhibitors, and gel conditions to resolve both species.
  • When performing immunohistochemistry (IHC-P), validate fixation protocols and antigen retrieval methods for your tissue type; positive controls should include autophagic-rich tissues such as heart, brain, or liver.
  • For ELISA applications, titrate the antibody to determine the optimal working concentration for either capture or detection setups, and evaluate cross-reactivity with closely related LC3 family members if needed.
  • Given the antibody's cross-reactivity with mouse and rat (as specified), it can be used in rodent model studies; however, verify species-specific performance in your experimental system.
