Biological Background: Major Vault Protein (MVP) Function and Localization
- MVP (Major vault protein), also known as lung resistance-related protein (LRP) or VAULT1, is the main structural component of vault particles—large ribonucleoprotein complexes implicated in cargo transport and signal transduction. Related references: PMID:40691181
- The protein oligomerizes to form a cage-like vault structure that can encapsulate minor vault components such as PARP4. Related references: PMID:40691181
- Vaults are believed to participate in nucleo-cytoplasmic transport, shuttling molecules between the nucleus and cytoplasm. Related references: PMID:15133037
- MVP negatively regulates interferon-gamma (IFNG)-mediated STAT1 signaling and downstream JAK activation. Related references: PMID:16418217
- MVP down-regulates SRC activity and MAP kinase signaling pathways. Related references: PMID:16441665
- Subcellularly, MVP localizes to the cytoplasm, nuclear pore complexes, and perinuclear region, consistent with its transport roles.
- MVP is expressed in most normal tissues, with higher levels in epithelial cells with secretory/excretory functions and in cells chronically exposed to xenobiotics; it is frequently overexpressed in multidrug-resistant cancer cells.
- Post-translational modifications include acetylation, phosphorylation, and ubiquitination, and the protein features multiple repeat domains.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (amino acids 1–260) of human MVP; consider this when interpreting band patterns or cross-reactivity in orthologs.
- For Western blot (WB), the predicted molecular weight is ~99 kDa; however, note that MVP can form high-molecular-weight complexes, so verify under reducing conditions.
- For immunohistochemistry (IHC-P), optimal dilution and antigen retrieval should be determined empirically for each tissue type.
- In ELISA, the antibody may be used as capture or detection reagent; validate pairing with appropriate secondary antibodies.
