- MAGED1, also known as NRAGE or DLXIN-1, is a member of the melanoma-associated antigen (MAGE) family, classified as a tumor antigen.
- It is involved in the apoptotic response after nerve growth factor (NGF) binding in neuronal cells, inhibits cell cycle progression, and facilitates NGFR-mediated apoptosis.
- The protein may act as a regulator of DLX family members and enhance the ubiquitin ligase activity of RING-type E3 ligases by stabilizing the E2 enzyme at the E3:substrate complex.
- MAGED1 plays a role in circadian rhythm regulation, likely as a co-regulator of RORA, modulating the expression of core clock genes such as BMAL1 and NFIL3 (induced) or NR1D1 (repressed).
- Subcellularly, MAGED1 localizes to the cytoplasm, cell membrane, and nucleus.
- It is ubiquitously expressed, including in bone marrow stromal cells from both multiple myeloma patients and healthy donors.
- The protein undergoes phosphorylation and alternative splicing, and participates in the ubiquitin conjugation pathway.

- The immunogen corresponds to the C-terminal region (amino acids 635–834), which is highly conserved across human, mouse, and rat; consider verifying specificity in your target tissue lysates.
- For Western blot, the predicted molecular weight is 86 kDa; validate the detected band pattern with appropriate positive and negative controls.
- In IHC-P, antigen retrieval conditions may need optimization to achieve optimal staining; consider testing citrate or EDTA-based protocols.
- As a polyclonal antibody, it may recognize multiple isoforms or post-translationally modified forms; confirm target identity with siRNA knockdown or overexpression if needed.
