Biological Background: MAGEA6 Function and Localization
- MAGEA6 (Melanoma-associated antigen 6), also designated Cancer/testis antigen 1.6 (CT1.6) or MAGE3B, is a member of the MAGE family of tumor antigens.
- Functions as an activator of ubiquitin ligase activity of RING-type E3 ubiquitin-protein ligases and acts as a repressor of autophagy. Related references: PMID:17942928 PMID:20864041 PMID:31267705
- Enhances ubiquitin ligase activity of TRIM28, thereby stimulating p53/TP53 ubiquitination; proposed to act through recruitment or stabilization of the Ubl-conjugating enzyme (E2) at the E3:substrate complex. Related references: PMID:17942928 PMID:20864041
- May play a role in tumor transformation or aspects of tumor progression; in vitro, it promotes cell viability in melanoma cell lines. Related references: PMID:17942928
- Expressed in a variety of tumors including melanoma, head and neck squamous cell carcinoma, lung carcinoma, and breast carcinoma, but not in normal tissues except for testes.
- Classified under tumor antigens and the ubiquitin conjugation pathway, reflecting its role in protein modification and cancer biology.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 75–314 of human MAGEA6; this region may be conserved across species, supporting detection in human and mouse (validated cross-reactivity).
- For Western blot (WB), consider using whole cell lysates from testis tissue or MAGEA6-positive tumor cell lines; the predicted molecular weight is ~35 kDa, though post-translational modifications may alter migration.
- For immunofluorescence/ICC, validate in cell lines known to express MAGEA6; appropriate fixation and permeabilization should be optimized.
- The polyclonal nature may provide robust signal but could result in detection of non-specific bands; include appropriate controls.
- ELISA applications can be developed using this antibody as a capture or detection reagent; pairing with a monoclonal detection antibody may improve specificity.
