Biological Background: MAGE-1/MAGE1A Function and Localization

• MAGE-1 (Melanoma-associated antigen 1) is a cancer/testis antigen encoded by the MAGEA1 gene, also known as CT1.1 or MAGE1.
• It may function as a transcriptional regulator by interacting with SNW1 and recruiting histone deacetylase HDAC1, and may inhibit NICD transactivation.
• The protein is expressed in the cytoplasm and nucleus.
• MAGE-1 is expressed in various tumors including melanoma, head and neck squamous cell carcinoma, lung carcinoma, and breast carcinoma, but is restricted to testis among normal tissues; it is absent in kidney tumors, leukemias, and lymphomas.
• It is recognized as a tumor antigen by autologous cytolytic T-lymphocytes.
• Post-translational modifications include phosphorylation, and it belongs to the MAGE family of proteins.

Experimental Guidance and Technical Tips

• The immunogen was a synthetic peptide corresponding to the region 200-309 of human MAGE-1. For Western blotting, expect a band around 34 kDa. Validate in relevant tumor cell lines or tissue samples.
• For ELISA, consider using recombinant MAGE-1 protein as coating antigen. Cross-reactivity with mouse indicates potential use in mouse xenograft studies.
• The antibody is rabbit monoclonal, offering high specificity. However, due to restricted expression, positive controls must express MAGE-1 (e.g., melanoma cell lines).
