- Myelin-associated glycoprotein (MAG), also designated Siglec-4a, is encoded by the MAG gene (GeneID: 4099) and exists as two major isoforms (isoform 1 and isoform 2) produced by alternative splicing.
- MAG functions as an adhesion molecule that mediates interactions between myelinating cells and neurons by binding to neuronal sialic acid-containing gangliosides and to the glycoproteins RTN4R and RTN4RL2.
- It is not essential for initial myelination but is critical for the maintenance of normal axon myelination and for preventing degeneration of myelinated axons in adults.
- MAG acts as a negative regulator of neurite outgrowth, inhibiting axonal extension through binding to RTN4R/RTN4RL2 or gangliosides, depending on the neuronal cell type.
- Subcellularly, MAG is localized to the cell membrane and membrane rafts of myelinating glial cells.
- Both isoforms are abundantly expressed in myelinated structures of the central and peripheral nervous systems, including oligodendroglia, periaxonal myelin, and Schmidt-Lanterman incisures.
