Biological Background: MAG Function and Localization

  • MAG (Myelin-associated glycoprotein), also known as Siglec-4a, is a type I transmembrane glycoprotein belonging to the immunoglobulin superfamily.
  • It mediates cell-cell adhesion between myelinating cells and neurons by binding to sialic acid-containing gangliosides and to the glycoproteins RTN4R and RTN4RL2.
  • Plays a crucial role in maintaining axon myelination and protecting motoneurons from apoptosis, particularly after injury.
  • Acts as a negative regulator of neurite outgrowth; in sensory neurons, this inhibition depends only partially on RTN4R, RTN4RL2, and gangliosides.
  • Preferentially binds to alpha-2,3-linked sialic acid and specific gangliosides such as Gt1b.
  • Localizes to the cell membrane and membrane rafts; in the nervous system, it is detected in periaxonal myelin and Schmidt-Lanterman incisures, with two isoforms (isoform 1 and isoform 2) expressed in both central and peripheral nervous system myelin (PubMed:6200494, PubMed:9495552). Experimental Guidance and Technical Tips

  • For Western blot (WB), expect a band around 69 kDa; consider using reducing conditions and whole cell lysates from neuronal or glial cell lines as positive controls.
