Biological Background: MAD1/MAD1L1 Function and Localization

  • MAD1/MAD1L1 (MAD1L1 gene) encodes mitotic spindle assembly checkpoint protein MAD1, also known as HsMAD1, hMAD1, and Tax-binding protein 181.
  • It is a core component of the spindle-assembly checkpoint (SAC) that delays anaphase onset until all chromosomes are properly aligned at the metaphase plate. Related references: PMID:10049595, PMID:20133940, PMID:29162720
  • At unattached kinetochores, MAD1 forms a heterotetrameric complex with closed MAD2L1 (C-MAD2) and catalyzes the conversion of open MAD2L1 (O-MAD2) to C-MAD2, amplifying the “wait anaphase” signal. Related references: PMID:29162720
  • MAD1 can also sequester MAD2L1 in the cytoplasm, impairing SAC function and promoting chromosomal instability, particularly in hepatocellular carcinomas.

  • Predicted molecular weight of MAD1 is ~83 kDa; a band near this size should be expected in Western blot. Consider using reducing conditions and appropriate loading controls.
  • For ELISA, the antibody may be used as a capture or detection reagent; optimize coating concentration and pairing with matched antibodies for sandwich ELISA development.
  • Cross-reactivity is reported for human and mouse; validate in other species if needed.
